Cold storage of Pacific oysters out of water: biometry, intervalval water and sensory assessment ArchiMer

Due to the important economic activity of oyster production in France, microalgal toxic events represent a significant constraint for shellfish farmers who face closures of bivalve production-sites. The frequency of closures of 3 weeks or less represents half of the total closures along the French coasts. Cold storage could be a simple and affordable solution for temporary and short-term storage before commercialisation. A stock of marketable Pacific oyster Crassostrea gigas was therefore stored during 22 days in non-immersed conditions at 3 degrees C with 100% humidity. As of 1985, Ifremer set up a selection programme for resistance to the protozoan Bonamia ostreae in European flat oysters (Ostrea edulis). Two selection lines, L85 and L89, from the third and second generation, respectively, were produced with 17 and 21 different families. The experimental design also included a mixed line (L85 x L89) of seven families and a control line of five families from Quiberon broodstock maintained without selection pressure.

A bulk production of phytoplankton (Skelelonema costatum) has been obtained in concrete tanks from saline groundwater. This was used in various concentrations to feed an intensive cultivation of the Manila clam (Ruditapes philippinarum) in ponds. No difference was observed in growth rates of molluscs, with food concentrations between 31 to 126 µg/l Chl a. However, the growth rates obtained from March to November with a food supplement were 1.5 times higher than those observed in the traditional, extensive cultivation. Summer mortality associated with juveniles of the oyster Crassostrea gigas is the result of a complex interaction between the host, pathogens and environment. Divergent selection based on summer survival have been applied to produce families with resistant and susceptible progeny. Here, we describe the use of suppression subtractive hybridization to generate 150 clones that were differentially regulated between resistant and susceptible F2 progeny. Spatial changes in the dietary regime of Ruditapes philippinarum from intertidal sediments of Arcachon Bay were depicted by analyzing stable isotope ratios in both its adductor muscles and potential trophic sources. Manila clams were collected from 50 sites in May to June 2006. A hydrobiological study coupled with the evolution of the production of cupped oysters Crassostrea gigas has been carried out from March 1990 to March 1991 on two oyster culture areas (North and South) of the Bay of Bourgneuf. - The northern area is characterized by an environment which is variable though time and composed of a high nutritive and mineral load. This high particular load (average value = 154 mg.l- 1) is linked to meteorological conditions and especially to the wind speed and direction. Un contrôle des performances de souches d'huîtres plates, Ostrea edulis, issues d'un plan de sélection a été réalisé d'avril à novembre 1995 en élevage intensif. Un débit d'eau de mer fixé à 4l/heure/huître et un apport nutritif (Skeletonema costatum) de 3.10(9) cellules/jour/huître semblent constituer un bon compromis pour satisfaire les exigences physiologiques de l'huître plate. Les lots d'huîtres résistantes ou tolérantes à Bonamia ostreae présentent de meilleures performances de croissance que les lots témoins. In this study, an original use of immersed microfiltration membranes (mean pore size equal to 0.2 mu m) is investigated for the total removal of toxic dinoflagellates from seawater. Using a membrane autopsy and fouling model approach and the use of fouling indexes (called Pore Blocking Index, Pore Constriction Index and Cake Filtration Index), three dinoflagellate suspensions (Heterocapsa triquetra, Alexandrium minutum and Prorocentrum lima) have been microfiltered in order to study the influence of micro-algal species and its concentrations (1,000 and 30,000 cells/mL) on filtration yield and membrane fouling mechanisms. Spatial and temporal variations in gill and palp size were studied during 1 year in naturally-settled populations of the Pacific oyster Crassostrea gigas, reciprocally transplanted between two contrasting sites located along a marked gradient of turbidity conditions. The variability of suspended particulate matter (SPM) and food particles, estimated by the concentration of chlorophyll-a, was measured with in situ water-quality probes. Shellfish industry is often realised in estuarine areas, subjected to pollutions due to agricultural activities. The harmful effects of pesticides on animals inhabiting these estuarine zones are poorly documented. Bivalve molluscs represent an interesting model because they are sedentary and they filter water intensively. Effects of pesticides on haemocytes functions were tested in Pacific oyster, Crassostrea gigas and analysed by flow cytometry. A combination of pesticide at environmental concentrations was tested in vivo. Animals used corresponded to two genetically selected populations produced in hatchery.

La filtration et la consommation d'oxygène d'Ostrea edulis ont été mesurées à différentes températures (10°-30°C) sur des individus de 5 à 120g de poids total (0,1 à 2,7g de poids sec) et hors période de gamétogénèse.
